Background information

The ELISpot is a common method for monitoring cellular immune responses to antigen stimulation. The general setup consists of a 96-well microtiter plate (strip or solid format), with a PVDF membrane at the bottom of each well. A capture antibody is immobilized onto the membrane to bind cytokines secreted by the antigen-stimulated cells. After applying an enzyme-conjugated detection antibody and following an enzymatic staining reaction, spots are revealed whereby one spot represents the unique cellular activity of antigen-specific cytokine-secreting cells at a single-cell level.

Comparative analysis of IFN-γ ELISpot kits

The T-Track® ELISpot kit human IFN-γ HiSpecificityPRO was compared to a competitor’s human IFN-γ ELISpot kit to measure T cell-mediated release of IFN-γ at a single-cell level (Figures 1-2). Per well, 100,000 freshly-prepared peripheral blood mononuclear cells (PBMCs) from three CMV-seronegative and three CMV-seropositive donors were used. For comparative analysis, stimulation of PBMCs was performed with JPT PepMix™ peptide pool pp65 for 20 h at 37°C and 5% CO2. Experimental procedures were performed according to the respective manufacturer‘s protocol. IFN-γ release was determined either in a one-step (Lophius) or a two-step detection system (Competitor).

In contrast to the competitor’s ELISpot kit (Figure 1D), no background spot was detected following pp65 stimulation of PBMCs from CMV-seronegative donors using Lophius’ ELISpot kit (Figure 1C), limiting the risk of false-positive results. Beside a reduced background and increased specificity, results obtained by the Lophius ELISpot kit are characterized by the detection of discrete, homogeneous spots from pp65-stimulated PBMCs from CMV-seropositive donors (Figure 1A), allowing accurate spot enumeration, unlike the competitor’s ELISpot read-out (Figure 1B).

Lophius’ IFN-γ HiSpecificityPRO ELISpot kit offers a highly specific and reliable assay with very low spot counts in unstimulated PBMCs from various CMV- and CMV+ donors (Figure 2A, green bars), as opposed to its competitor’s IFN-γ ELISpot kit (Figure 2A, grey bars). Following pp65 stimulation (Figure 2B), Lophius’ IFN-γ HiSpecificityPRO ELISpot kit demonstrated higher specificity and comparable sensitivity (green bars) over its competitor kit (grey bars).
